The translation and processing of secretory proteins starts in the cytosol and then proceeds as follows:
cytosol → {{c1::rough endoplasmic reticulum}} → {{c1::golgi apparatus}} → {{c1::secretory vesicles}} → {{c1::plasma membrane}}

- Translation of mRNA to proteins begins in the cytosol. From there, proteins meant for cellular use travel to their intracellular destination.
- However, proteins intended for the secretory pathway have N-terminal signal sequences that causes them to be transferred to the rough endoplasmic reticulum (RER)
- In the RER, post-translational modifications such as glycosylation, disulfide bond formation, phosphorylation, and protein cleavage can occur.
- The RER then packages proteins into vesicles and sends them to the golgi aparatus for further processing (retrograde processing)
- Finally, the golgi apparatus packages proteins into secretory vesicles which fuse with the plasma membrane and releases the contents outside of the cell
